However, its specific regulatory function, its ligand, or the operator sites have not yet been established. Based on its sequence similarity to the HutC proteins, PhnF is predicted to regulate the expression of the phn genes and respond to alkylphosphonates. PhnF from Escherichia coli, which is the focus of this article, belongs to the phn operon that is involved in transport and biodegradation of phosphonates, (Pn)-compounds having carbon–phosphorous (C–P) bonds ( Metcalf and Wanner 1993). 1994), the trehalose operon repressor TreR from Bacillus subtilis that is inhibited by trehalose-6-phosphate ( Schock and Dahl 1996), and a number of proteins (KorSA, KorA, and TraR) that are repressors of the genes involved in conjugative plasmid transfer in Streptomyces species ( Kendall and Cohen 1988 Hagege et al. Other characterized members of the HutC family include FarR from Escherichia coli, which regulates the expression of citric acid cycle genes and responds to long-chain fatty acids ( Quail et al. It had been named after the HutC regulator from Pseudomonas putida, which represses the expression of histidine utilization genes and is, in turn, inactivated by the binding of uroconate ( Allison and Phillips 1990). The HutC subfamily represents more then 30% of all GntR members. 2001), no structural information is available for the other three subfamilies of GntR-like regulators. While the structure of FadR alone and in complex with its effector and operator DNA has been recently determined ( van Aalten et al. According to this analysis, the GntR family has been divided into four major subfamilies, FadR, HutC, MocR, and YtrA, where each subfamily is categorized by a specific type of the C-terminal domain. Recent sequence analysis of the GntR proteins revealed the presence of several distinct groups with different types of the C-terminal signaling domains ( Rigali et al. This output domain is coupled with the C-terminal signaling domain responding to a range of stimuli in the form of different small molecules. The proteins in the GntR family share a characteristic version of the N-terminal winged helix-turn-helix (wHTH) DNA-binding domain. This is true for the GntR family of transcriptional regulators, which gathers close to 2000 members in both bacterial and archael genomes ( Haydon and Guest 1991 Rigali et al. With extensive sequence similarity in the DNA-binding domain, these large families often contain proteins with different types of the effector domains fused to similar DNA-binding domains ( Rosinski and Atchley 1999 Perez-Rueda and Collado-Vides 2000). 1994), GntR ( Haydon and Guest 1991), and AraC ( Gallegos et al. Some of the largest such families ( Perez-Rueda et al. Based on sequence similarity, predominantly in the DNA binding domain one-component systems identified in sequenced genomes are assembled into protein families usually named after the best-characterized member. Signal transduction in prokaryotes is dominated by such systems, where small molecule-binding domains constitute the majority of the input domains and helix-turn-helix (HTH) DNA-binding domains are the most common output domains ( Ulrich et al. The structural comparison of the C-PhnF and UbiC proteins allows us to propose that a similar site in the PhnF structure is adapted for effector binding.Ī protein containing a sensor input domain and a functional output domain is described as a one-component system ( Ulrich et al. C-PhnF shares strong structural similarity with the chorismate lyase fold, which features a buried active site locked behind two helix-turn-helix loops. Members of this family share similar N-terminal DNA-binding domains, but are divided into four subfamilies according to their heterogenic C-terminal domains, which are involved in effector binding and oligomerization. The PhnF protein belongs to the HutC subfamily of the large GntR transcriptional regulator family. The crystal structure of Escherichia coli PhnF C-terminal domain (C-PhnF) was solved at 1.7 Å resolution by the single wavelength anomalous dispersion (SAD) method. ![]() While some individuals may perceive themselves susceptible to the media impacts, some others may believe that they are resistant to the content of media. Perceived pressure from media can be entangled with the perceived effects of media on self. Ī sociocultural perspective argues that perceived pressure from media (both traditional and digital media) to conform to the published ideals leads to the internalization of unrealistic body standards, upward comparison, unhealthy appearance schemas and appearance preoccupation. Fitspiration became popular as the channel to promote healthy living focusing on diet and exercise, but it has eventually developed into a platform mainly hosting the images of thin bodies for females and muscular or hypermuscular bodies for males, which are associated with an ideal image for many individuals. In the increasingly digital world and the proliferation of social networking sites (SNS), Instagram has become one of the platforms for everyone to share appearance ideals using a hashtag, #fitspiration (a blended word combining ‘fitness’ and ‘inspiration’). ![]() ![]() These results accentuate the value of incorporating a potential mediator and moderator into the direct relationship between internal locus of control and perceived effect of media ideals on self to provide an enhanced understanding of this process and offer practical insights about the protective role of media literacy and negative effects of high BMI.įor years, the media has created a delusion that men and women should fit the narrowly defined standards of ideal societal appearance. However, BMI moderates the mediated relationship such that the negative relationship between internal locus of control and the perceived effect of fitspiration images through media literacy does not exert an effect on those with high BMI. Results show that internal locus of control is negatively associated with the perceived impact of fitspiration images on self, mediated by media literacy. The data analysis was performed using SPSS PROCESS macro. A sample of 321 Malaysian university students who were fitspiration viewers filled out a self-report questionnaire assessing internal locus of control, media literacy, perceived effect of fitspiration images on self, body satisfaction and BMI. This study also examined the importance of body mass index (BMI) as a moderating factor in the mediated model, where higher BMIs could weaken the relationship between internal locus of control and the perceived effect of fitspiration images mediated through media literacy. The present study investigated the relationship between internal locus of control and the perceived impact of Instagram fitspiration images on the self with media literacy as the mediating role in this relationship. You probably will notice a lag when you change channels. After all, how often are you watching the same TV program from two different sources at once? But it does raise the curious possibility that while you're watching a big basketball game on ESPN you could hear raucous cheering from your neighbors' house only to discover- two minutes later-that the fuss was abut a mind-blowing, buzzer-beating, game-winning three-pointer. Truth be told, most subscribers to Sling TV would never notice the delay. We expect the perceived delay will be significantly reduced over time." ![]() Since Sling TV is a new service, we have chosen to be conservative in our signal delay to allow for congestion or other potential Internet propagation delays. They all have delay from the time of content was captured and produced to the time a viewer sees the signal. "Television signals of all types are never truly live. We asked the Sling TV spokesperson about the delay, and got this explanation: We even tried it at the home of a staff editor who subscribes to Dish Network's satellite service and found Sling TV to be running about a minute behind the company's own feed from space. Side by side, we found that Sling TV was running one to two minutes behind the programming on Optimum cable. The most striking difference we saw between the cable and Sling TV feeds was one of latency. The organizational logic makes a lot more sense than assigning arbitrary numbers to channels, but its difficult to know how it would scale beyond the dozen or so channels currently on Sling TV. On Sling TV, channels have no number and instead are grouped by type (Sports, Family, News, etc.). Navigating Sling TV is a bit of a departure from the standard-issue cable and satellite service numerical channel guide. If you want to watch Sling TV on your television, you might need to spend $40 to $100 for a streaming media player, but that's still a significant savings over the multiyear cost of a DVR or a cable box. It should also work on Android and Apple iOS devices. At launch, Sling TV should work with streaming media players from Amazon and Roku as well as with the Xbox One, Google Nexus Player, and smart TVs from LG and Samsung. Like many other streaming video services, Sling TV works through an app that you can install on multiple devices. No launch date for Sling TV has been announced, but the company assured us it's coming soon. Premium channels, such as HBO, Cinemax, and Showtime, are also unavailable on Sling TV. Soon it becomes hard to tell which characters are actors in his play from which characters are actors in the film attempting to live their lives in peace. Written and directed by Charlie Kaufman, Synecdoche, New York blurs Caden’s imagination with the reality of the world around him. After facing a series of awful and unfortunate events, he becomes obsessed with a play he is producing and devotes his entire life to making it his masterpiece. Synecdoche, New York is a reverse application of the cave analogy, because it follows Caden Cotard (Phillip Seymour Hoffman), a theater director in Schenectady, as he leaves reality and enters the cave. But, no Hitchcock film is truly a Hitchcock film without a murder, so anticipation builds and the film builds up to a dramatic climax. In fact, of this list, this is the most visually allusive to the allegory of the cave. Inside the cave, Hitchcock gives viewers an eerie remake of the cave allegory, as the camera follows the shadows of those in the boat just through a cave. Bruce then stalks Guy’s wife at a carnival and eventually climbs onto a boat she and others are riding to an island. In what can only be described as a perfect coincidence, he runs into Bruce Anthony (Robert Walker) on a train, who convinces him that he is the perfect person to commit such a murder. Thanks to a speech Emmet gives, Lord Business reforms simultaneously while the boy’s father realizes that gluing the pieces together inhibits his son’s creativity, and so both worlds are saved as the “Piece of Resistance” is used to cap the “Kragle.” Thus, by turning to the higher realm, Emmet is able to save Lego World. The boy’s father has been gluing the pieces together to keep them the way he envisioned Lego World, to his son’s chagrin. When the real world is revealed to viewers, we realize a boy has been playing with Lego pieces and that “The Kragle” is actually Krazy Glue with letters missing from the tube. Analyzed at this level, the film seems a bit cliché, but there is much more depth. After being captured and watching his friend die, Emmet escapes and rallies the people of Lego World to fight against Lord Business. I limit this list to one film per director, though many of these directors have several great platonic films. In what is to follow, I list 10 great films that feature Platonic cave allegories. Nearly all philosophy is indebted to Plato in one way or another, so unsurprisingly many films allude to him and to his cave allegory in particular. Sound familiar?
